Bioinformatics analysis for drug repositioning

Abstract
Objectives: Development of a new medication is not an easy task, as it is not only financially costly but also very time consuming without any guarantee of success. Information for medications is well-accumulated due to technological advances, especially IT technology. This information has allowed research to be conducted regarding the repositioning of drugs for the new uses based on the side effect information of existing medications. We examine effectiveness and safety of off-label use of existing drugs for rare diseases using Bioinformatics techniques. Some drugs used for the treatment of certain disease will be used for other diseases with common shared genes.

Methods: Gene information associated with the diseases was collected from Genotator. Our research paired authorized indication and off-label use of existing medication. We found shared genes in two pairs of diseases. Protein-protein interaction networks consisting of shared genes were produced using STRING. The target proteins, side effects, interacting agents, and structural information of each drug were collected from DrugBank. Moreover, the drug-similarity network was generated using the Cytoscape.

Results: The research could confirm the association of drugs to be used between the diseases in a pair. Drugs targeting the shared gene are already used for both diseases. Therefore, we can recommend a safe and effective medicine for rare disease using this drug-similarity network.

Conclusion: We can confirm a potential for drug repositioning by studying common genes. Through this research, we were able to investigate the possibility of the use of Bioinformatics in finding new utilization of existing drugs.
